IGF-1 DES (Des(1-3)IGF-1) is a naturally occurring truncated form of Insulin-like Growth Factor-1, produced by the removal of the first three N-terminal amino acids (glutamate, proline, and threonine). This structural modification significantly reduces its binding affinity to IGF-binding proteins (IGFBPs), which in turn increases the concentration of bioavailable, free peptide at the tissue level. As a result, IGF-1 DES demonstrates approximately 10-fold greater potency than standard IGF-1 in in vitro receptor binding assays.

In research settings, IGF-1 DES has been investigated for its pronounced effects on cellular proliferation, differentiation, and survival pathways — primarily through activation of the IGF-1 receptor (IGF-1R) and downstream PI3K/Akt and MAPK/ERK signalling cascades. Studies have explored its potential applications in muscle cell hypertrophy, neuroprotection, wound healing, and metabolic regulation.
