PNC-27 is a synthetic peptide derived from the MDM-2-binding domain of the p53 tumour suppressor protein, combined with a membrane-targeting sequence. Unlike conventional cytotoxic agents, PNC-27 is proposed to exert its effects by selectively binding to HDM-2 (the human homologue of MDM-2) expressed aberrantly on the membranes of cancer cells, leading to the formation of pores and subsequent tumour cell lysis. This membrane-disrupting mechanism is of significant interest to oncology researchers, as it suggests a degree of selectivity towards malignant cells whilst leaving normal cells relatively unaffected.
PNC-27 has been investigated in preclinical research across a range of cancer cell lines, including pancreatic, breast, and leukaemia models, demonstrating cytotoxic activity in vitro.
