Dermorphin is a naturally occurring heptapeptide first isolated from the skin of the South American frog Phyllomedusa sauvagei. It is widely regarded as one of the most potent endogenous opioid peptides identified to date, exhibiting a binding affinity for mu-opioid receptors (MOR) estimated to be 30–40 times greater than that of morphine on a molar basis. Its unusual D-amino acid configuration at position 2 (D-Ala) confers exceptional resistance to enzymatic degradation, making it a valuable research tool for studying opioid receptor pharmacology and ligand–receptor interactions.
In preclinical research settings, dermorphin has been employed to investigate analgesic signalling pathways, opioid receptor selectivity, and the neurobiological mechanisms underpinning pain modulation. It has also served as a scaffold for the development of novel opioid-based therapeutic candidates and receptor-selective analogues.
