ACE-031 (also known as ACVR2B-Fc) is a recombinant fusion protein comprising the extracellular domain of activin receptor type IIB (ActRIIB) linked to a human IgG1 Fc region. By acting as a ligand trap, ACE-031 competitively binds circulating negative regulators of muscle growth — most notably myostatin (GDF-8), activin A, GDF-11, and BMP-9 — preventing them from engaging endogenous ActRIIB receptors. This mechanism effectively suppresses the ActRIIB-mediated signalling cascade that limits skeletal muscle hypertrophy, making it a subject of considerable interest in preclinical musculoskeletal research.
In research settings, ACE-031 has been investigated for its potential to increase lean muscle mass and improve muscle function in models of muscular dystrophy, cachexia, sarcopaenia, and other muscle-wasting conditions. Studies have also explored its influence on bone density and metabolic parameters, given the cross-talk between the myostatin/activin pathway and bone remodelling.
