FOXO4-DRI, also known by its experimental designation Proxofim, is a retro-inverso peptide analogue engineered to interfere with the interaction between the transcription factor FOXO4 and the tumour suppressor protein p53. In senescent cells, FOXO4 sequesters p53 within the nucleus to prevent apoptosis, allowing these dysfunctional cells to persist and secrete pro-inflammatory factors collectively termed the senescence-associated secretory phenotype (SASP). FOXO4-DRI competitively disrupts this interaction, thereby restoring p53’s capacity to translocate to the mitochondria and initiate apoptosis selectively in senescent cells — a process termed senolysis.
This peptide has attracted considerable interest in preclinical ageing research, particularly in models examining tissue rejuvenation, age-related frailty, fibrosis, chemotherapy-induced senescence, and chronic inflammation. Its high selectivity for senescent over healthy proliferating cells makes it a valuable tool for studying targeted senescent cell clearance without broad cytotoxic effects.
