Orexin B, also known as Hypocretin-2, is a 28-amino acid neuropeptide produced by neurones in the lateral hypothalamus. It is one of two orexin peptides (alongside Orexin A) derived from the prepro-orexin precursor protein and plays a critical role in the regulation of sleep-wake cycles, arousal, appetite, and energy homeostasis.
Orexin B exerts its effects primarily through the OX2 receptor (orexin receptor type 2), with lower affinity for OX1 receptors compared to Orexin A. This receptor selectivity makes Orexin B a valuable tool in research aimed at distinguishing the distinct physiological roles of each receptor subtype. Dysregulation of the orexin system has been implicated in narcolepsy, insomnia, depression, and metabolic disorders, making Orexin B a subject of considerable scientific interest.
