Melanotan 1 (MT-1), also known as afamelanotide, is a synthetic linear analogue of the endogenous peptide alpha-melanocyte-stimulating hormone (α-MSH). It was developed to investigate the melanocortin receptor pathway, particularly its interaction with the melanocortin 1 receptor (MC1R), which plays a central role in regulating eumelanin production and skin pigmentation.
In research settings, MT-1 has been extensively studied for its ability to stimulate melanogenesis — the biological process responsible for melanin synthesis in melanocytes. By binding to and activating MC1R, MT-1 triggers a downstream signalling cascade involving cyclic AMP (cAMP), leading to increased tyrosinase activity and enhanced eumelanin output. This mechanism has made it a subject of interest in photoprotection research, erythropoietic protoporphyria (EPP) models, and dermatological studies.
Beyond pigmentation, MT-1 has attracted research interest in areas including immune modulation, anti-inflammatory responses, and neuroprotection, given the broad distribution of melanocortin receptors throughout peripheral and central tissues.
