PE-22-28 is a spirocyclic, stapled synthetic peptide derived from spadin, a naturally occurring fragment of the sortilin propeptide. It functions as a selective antagonist of the TREK-1 (TWIK-related potassium channel 1) channel — a two-pore domain background potassium channel implicated in the regulation of mood, neuronal excitability, and the physiological response to anaesthetics and neuroprotective stimuli.
Compared to its parent peptide spadin, PE-22-28 has been engineered for significantly improved metabolic stability and bioavailability, making it a more practical and reproducible tool for in vitro and in vivo research applications. Preclinical studies have explored its rapid-onset antidepressant-like effects, with evidence suggesting action within timeframes considerably shorter than conventional monoaminergic antidepressants.
Research has additionally investigated PE-22-28 in the context of hippocampal neurogenesis, synaptic plasticity, and cognitive function, positioning it as a valuable compound in the broader study of psychiatric and neurodegenerative conditions.
