PNC-27 is a synthetic peptide derived from the transactivation domain of the p53 tumour suppressor protein, incorporating an HDM-2 binding domain fused with a transmembrane-penetrating sequence. This unique structural design has made it a subject of considerable interest in oncology research. Unlike many conventional anticancer research compounds, PNC-27 is believed to exert its effects by directly targeting and disrupting the plasma membranes of cancer cells whilst leaving normal, healthy cells relatively unaffected — a mechanism thought to relate to the overexpression of HDM-2 (the human homologue of MDM2) on tumour cell membranes.
In laboratory studies, PNC-27 has been investigated for its selective cytotoxic activity across a range of cancer cell lines, including pancreatic, breast, and leukaemic cell models.
