Retatrutide (also known as LY3437943) is a novel triple receptor agonist that simultaneously targets glucagon-like peptide-1 (GLP-1), glucose-dependent insulinotropic polypeptide (GIP), and glucagon receptors. This triagonist mechanism distinguishes it from dual agonists such as tirzepatide, and has made it a subject of significant interest in metabolic research.
In preclinical and clinical studies, retatrutide has demonstrated substantial effects on body weight reduction, glycaemic regulation, and lipid metabolism. The glucagon receptor component is thought to enhance energy expenditure and hepatic fat reduction, whilst the GLP-1 and GIP components contribute to appetite suppression and improved insulin sensitivity. Phase 2 clinical trial data has shown some of the most pronounced weight reduction results recorded among incretin-based investigational compounds to date.
