SS-31, also known as Elamipretide or MTP-131, is a synthetic aromatic-cationic tetrapeptide comprising the sequence D-Arg-dimethylTyr-Lys-Phe-NH₂. It is specifically designed to target and associate with the inner mitochondrial membrane, where it interacts with cardiolipin – a phospholipid essential for the structural integrity and functional efficiency of mitochondrial electron transport complexes.
In preclinical research settings, SS-31 has been investigated for its capacity to stabilise cardiolipin, reduce mitochondrial reactive oxygen species (ROS) production, and preserve ATP synthesis under conditions of cellular stress. These properties have made it a subject of considerable interest across multiple research disciplines, including studies into ischaemia-reperfusion injury, age-related mitochondrial dysfunction, neurodegenerative conditions, and cardiomyopathy models.
SS-31 has also featured in research exploring skeletal muscle energetics, renal protection, and metabolic decline associated with ageing. Its selective localisation to the inner mitochondrial membrane, rather than the cytoplasm, distinguishes it from broader antioxidant compounds and gives it a precise mechanism of action relevant to mitochondrial biology research.
