Vasoactive Intestinal Peptide (VIP) is a 28-amino acid neuropeptide found throughout the central and peripheral nervous systems, as well as in immune and endocrine tissues. It exerts its effects primarily through binding to VPAC1 and VPAC2 receptors, activating adenylyl cyclase and elevating intracellular cAMP levels, which mediates its broad range of biological actions.
In preclinical research, VIP has been extensively studied for its potent anti-inflammatory and immunomodulatory properties. It is known to suppress pro-inflammatory cytokines such as TNF-α, IL-6, and IL-12 whilst promoting regulatory T-cell activity, making it a subject of significant interest in autoimmune and inflammatory disease research. VIP also demonstrates vasodilatory effects across multiple vascular beds, bronchodilatory activity in pulmonary models, and neuroprotective properties in neurological studies.
Additionally, VIP has been investigated in the context of circadian rhythm regulation, gut motility, and neuroendocrine signalling, reflecting its diverse physiological roles.
