Dermorphin is a naturally occurring heptapeptide originally isolated from the skin secretions of South American frogs of the genus Phyllomedusa. It is one of the most potent naturally derived opioid peptides known, exhibiting exceptionally high affinity and selectivity for mu-opioid receptors (MOR). Notably, dermorphin contains a D-alanine residue at position 2, which confers resistance to enzymatic degradation and contributes significantly to its prolonged biological activity compared to endogenous opioid peptides.
In research settings, dermorphin is widely utilised as a pharmacological tool to investigate mu-opioid receptor binding kinetics, signal transduction pathways, and the neurophysiology of pain and analgesia. Its unique structural features make it valuable for structure–activity relationship (SAR) studies aimed at developing novel opioid analgesics with improved receptor selectivity and reduced side-effect profiles.
