Kisspeptin-10 is a biologically active ten-amino acid fragment derived from the larger kisspeptin family of neuropeptides, encoded by the KISS1 gene. Also referred to as metastin (45–54), it represents the C-terminal decapeptide sequence responsible for the majority of kisspeptin’s biological activity. Kisspeptin-10 exerts its effects by binding to the kisspeptin receptor (KISS1R, formerly known as GPR54), a G protein-coupled receptor expressed predominantly in hypothalamic neurons. Activation of KISS1R potently stimulates the release of gonadotropin-releasing hormone (GnRH), triggering downstream secretion of luteinising hormone (LH) and follicle-stimulating hormone (FSH) from the pituitary gland, making it a central regulator of the hypothalamic–pituitary–gonadal (HPG) axis.
In preclinical research, Kisspeptin-10 has been extensively studied in models of reproductive physiology, puberty onset, fertility, and conditions such as hypogonadotropic hypogonadism. It is also investigated in the context of tumour metastasis suppression, appetite regulation, and glucose homeostasis.
