MOTS-c (Mitochondrial Open Reading Frame of the Twelve S rRNA-c) is a 16-amino acid mitochondrial-derived peptide encoded within the 12S ribosomal RNA gene of the mitochondrial genome. Unlike conventional nuclear-encoded peptides, MOTS-c represents a unique class of mitochondrial peptides that translocate to the nucleus in response to metabolic stress, where it regulates adaptive gene expression.
Mechanism of Action: MOTS-c primarily activates the AMPK (AMP-activated protein kinase) pathway, a central regulator of cellular energy homeostasis. It inhibits the folate cycle and de novo purine synthesis, leading to AICAR accumulation and downstream AMPK activation. This results in enhanced glucose uptake, improved insulin sensitivity, and increased fatty acid oxidation at the cellular level.
