P21, also referred to as P021, is a tetrapeptide fragment derived from the ciliary neurotrophic factor (CNTF) binding site, specifically designed to mimic the neurotrophic activity of CNTF without engaging the full cytokine receptor complex. This small peptide has attracted considerable research interest due to its ability to penetrate the blood-brain barrier and modulate key neurotrophin signalling cascades.
In preclinical research settings, P21 has been investigated for its capacity to upregulate brain-derived neurotrophic factor (BDNF) expression, promote hippocampal neurogenesis, and support synaptic plasticity. Studies in rodent models have explored its potential relevance to cognitive decline, age-related neurodegeneration, and conditions associated with reduced neurotrophic signalling, including models of Alzheimer’s disease and Down syndrome.
P21 is also noted in research for its potential to inhibit tau hyperphosphorylation, a hallmark pathological feature of several neurodegenerative conditions, suggesting possible applications in tauopathy research.
